If you have 6 people who can rotate driving and they can all stand being in the car that long, you shouldn't have to stop for an overnight until you get to where you are going.

Switch drivers at least every gas stop. To keep fresh, I would change drivers every 3 hours or less. Yes, each one can drive longer than 3 hours, but it's safer to change more often since you have the drivers.
